Abstract: As described in a previous paper, with a dihydrotestosterone affinity Sepharose 4B column the authors isolated and purified the transformed type androgen receptor in high ionic strength from the rat testis. Now, with the same affinity column we isolated the nontransformed type androgen receptor in the presence of sodium molybdate from the rat testis. The molecular weight of the receptor estimated by gel chromatography was about 300,000 daltons or more. In a histocytochemical electron microscopic study, the colloidal gold-labeled nontransformed androgen receptor had an affinity to nuclear proteins which might be constituents of the active chromatin in a Sertoli cell nuclei. The DNase treatment indicated that actin is not the target of the binding of this type of the receptor. The nontransformed androgen receptor may localize in an active chromatin region as well as the transformed androgen receptor and regulate their hormone responsive gene in higher order chromatin structures.
