Abstract: The effects of OK-432-induced activated killer cell on the prevention of metachronous liver metastases were studied in mice. Colon 26 cells (5×10⁴) were inoculated into the portal vein of female BALB/c mice. On day 14 after inoculation, the mice showed 114±70 (mean±SD) nodules of liver metastasis. The median survival time was 27 days. We used this model for experimental adoptive immunotherapy of OK-432-induced activated killer cell (OK-AK cell). The mice were divided into two groups : one was the control group that was inoculated with colon 26 (5×10⁴) with untreated non-adherent splenocyte (2×10⁶) : the other was the OK-AK group that was inoculated with colon 26 cell (5×10⁴) with OK-AK cell (2×10⁶). The number of metastatic colonies and wet hepatic weight was significantly reduced in the OK-AK group (32.8±8.1, 0.98±0.07 g) compared with the control group (171.0±37.3, 1.23±0.23 g). Also the survival time of mice was significantly improved. These results demonstrate that perioperative adoptive immunotherapy with OK-432-induced activated killer cells reduced the development of liver metastases.
