drug, were examined, using conventional microelectrode technique and fura-2 Ca²⁺-sensitive fluorescent dye. Amiloride (50 μM) depressed the action potential amplitude by 8.2±1.4% (n=8, P<0.05) and the maximum rate of depolarization by 16.2±2.0% (n=8, P<0.01). In addition, the action potential duration was prolonged by 26.7±3.4% (n=6, P<0.05) at 30 μM, and the resting potential was depolarized by 11.8±1.7% (n=6, P<0.05) at 0.5 mM amiloride. In contrast, amiloride (0.5 to 1 mM) significantly increased the
contractile force by 8 to 30% (n=8), but tended to decrease it at lower concentrations (30 μM to 0.1 mM). The positive inotropic effect was not affected by propranolol (0.1 μM), a β-adrenoceptor blocker. In fura-2 loaded ventricular myocytes, amiloride (1 mM) initially elevated cellular Ca²⁺ level ([Ca]₁) by 24.5±2.9% (n=6, P<0.01), and during the application, the [Ca]₁ level declined. These results indicate that amiloride possesses complex cardiac (protective) actions : electrical inhibitory and mechanical stimulatory actions, accompanied with the elevation of cellular Ca²⁺ concentration.

Plasma AVP level and osmolality
(OSM) were measured before and after oral water loading in 14 normal volunteers, 20 non-medicated and 27 medicated schizophrenic patients with psychotropic drugs. Water loading test was performed twice, before and during medication, in the same 13 schizophrenic patients. 1. Simple regression analysis did not show significant differences in AVP secretion manner between the control and non-medicated groups. The sensitivity of AVP secretion response to osmolality in the medicated group was lower than that in the normal control group, suggesting that the psychotropic drug was a major factor in the change of AVP secretion manner. 2. In the medicated patients the basal levels of plasma AVP and ΔAVP/ΔOSM were lower than those in the same and non-medicated patients. 3. Multiple regression analysis in the non-medicated patients revealed associations of the Brief Psychiatric Rating Scale scores to their basal levels of AVP. AVP levels were
related positively to acute symptoms and negatively to conceptual disorders in the patients.

A 61-year-old woman visited the hospital because of tumor in the upper outer quadrant of the right breast. Palpation, an ultrasonography and mammography suggested breast cancer. Needle
aspiration biopsy of the mass yielded tumor cells, so a standard radical mastectomy was
performed. The pathological specimen revealed malignant lymphoma, diffused type, medium sized cell, B cell type on LSG classification. Postoperative examination disclosed no abnormal findings and so it was diagnosed as primary malignant lymphoma of the breast.

Serum amylase was very high, in spite of low level of urine amylase and normal value of serum lipase. The abnormal molecular size of amylase was detected by electrophoresis. Thin layer chromatography and immunofixation technique showed that the amylase binding molecule was immunoglobulin (IgGAκλ type). Both the pathogenesis and clinical significance of the association of these disorders in this patient are discussed.

HISTOPATHOLOGICAL STUDY OF THE URINARY BLADDER EPITHELIUM IN AGED DOGS常深, 邦彦urinary bladder epitheliumaged changesbeagle dogsApparently normal urinary bladder of beagle dogs of both sexes were examined histopathologically. Materials were obtained from fifty autopsied aged beagle dogs and six autopsied young beagle dogs. Changes of urinary bladder epithelium were classified into five types : Brunn's nest, squamous metaplasia, lymphocytic infiltration, epithelial hyperplasia, and atypia. Brunn's nests in aged dogs were found at all ages and in both sexes. Squamous metaplasia in aged dogs was found in 25.0 per cent of males and 50.0 per cent of females. Scattered lymphocytic infiltration in aged dogs could be observed in 18.0 per cent of both sexes. Epithelial hyperplasia and atypia in aged dogs were found in 82.0 per cent and 28.0 per cent of cases, respectively ; however, no difference between sexes was found. Sites of predilection for Brunn's nests were the trigone and anterior wall ; however, those for other types of lesion were not found. As for young dogs, Brunn's nests were found in 66.7 per cent of both sexes, and sites of predilection were the trigone and anterior wall ; the other lesions were not observed. These chronic changes of urinary bladder epithelium in aged beagle dogs should be considered in the bladder carcinogenesis
and natural history of bladder tumor in dogs.

All rats were given 0.05% BBN in drinking water for the initial 8 weeks of the experiment. Chemotherapeutic agents evaluated in this investigation were administered as single treatments during the subsequent 12 weeks and rats were sacrificed after 20 weeks of the experiment. The investigation was divided into 3 experimental groups according to animal strains and administration methods of chemotherapeutic agents : (1) intraperitoneal administration in male Wistar rats ; (2) oral administration in male Fischer 344 rats ; and (3) intraperitoneal administration in male Fischer 344 rats. In experimental group (1), 5-fluorouracil (5-FU), 1-(2-tetrahydrofuryl)-5-fluorouracil (FT-207), carboquone (CQ), vincristine (VCR) and cis-diamminedichloroplatinum (CDDP) were effective in suppressing the incidence of bladder tumor. On the other hand, doxorubicin (DXR), mitomycin-C (MMC), neocarcinostatine (NCS), cyclophosphamide (CPM) and bleomycin (BLM) were not effective. In experimental group (2), both UFT, 4:1 mixture of uracil and tegafur, and 1-hexylcarbamoyl-5-fluorouracil (HCFU) suppressed the incidence of bladder tumor. In experimental group (3), N, N', N"-triethylene thiophosphoramide (thiotepa) also suppressed the tumor incidence. These results indicate that 5-FU analogue, CQ, VCR, CDDP and thiotepa may be clinically effective for prophylactic treatment for superficial bladder cancer. In addition, continued use of this murine bladder model is recommended for future screening of chemotherapeutic agents for superficial bladder cancer.奈良医学会2011-12-08T04:37:24Z2011-12-08T04:37:24Z1993-08-31ArticleDepartmental Bulletin Paper1507217 bytesapplication/pdf奈良医学雑誌 Vol.44 No.4 p.236-2490469555013450069http://hdl.handle.net/10564/179404695550AN00180974奈良医学雑誌444236249jpnpublisher

The first study : A total of 130 evaluable cases were randomized into 3 groups : (1) intravesical instillation of doxorubicin (20 mg/40 ml, 30 mg/30 ml), mitomycin C (20 mg/40 ml), or thiotepa (30 mg/30 ml) ; (2) Non-instillation treatment with etretinate or tegafur ; and (3) controls. All agents were administered for 2 years. Significant suppression of recurrence was observed in stage T1 or grade 2 disease treated with prophylactic instillation administered for the first 24 months of the 48-month observation period. The second study : A total of 140 evaluable cases were randomized into 3 groups : (1) intravesical
instillation of thiotepa (30 mg/30 ml) ; (2) oral administration of UFT ; and (3) controls. All agents were administered for 2 years. Significant suppression of recurrence was observed in stage T1, grade 2 or multiple cases with prophylactic instillation of thiotepa for the first 24 months of the 36-month observation period. These 2 studies indicate that bladder instillation therapy of DXR, MMC and thiotepa had some prophylactic effects after TURBt in patients with stage T1, grade 2 and multiple superficial badder cancer.

Accordingly, we attempted to ascertain whether it was true or not, using another methodologies. Result : The meaning of the span of life would be considered as the age which is suited (or proper) at the top of the mountain formed within the range of 70 to 100 years of age. The span of life of the Japanese has been advancing and retreating for the past 17 years in Japan.
